When This Medicine Should Not Be Used:

This medicine is not right for everyone. Do not use it if you had an allergic reaction to capivasertib.

How to Use This Medicine:

Tablet

  • Take your medicine as directed. Your dose may need to be changed several times to find what works best for you. Take the medicine at about the same time each day.
  • Swallow the tablet whole. Do not chew, crush, or split it. Do not take this medicine if it is broken, cracked, or damaged.
  • Read and follow the patient instructions that come with this medicine. Talk to your doctor or pharmacist if you have any questions.
  • For women who have not reached menopause or are just starting menopause, your doctor may also give you luteinizing hormone-releasing hormone medicine together with this medicine.
  • Missed dose: If you miss a dose and it is within 4 hours of the usual time, take it as soon as you can. Then go back to your regular schedule. If you miss a dose and it is more than 4 hours of your usual time, skip the missed dose and go back to your regular dosing schedule. Do not take extra medicine to make up for a missed dose.
  • If you vomit after you take this medicine, do not take another dose. Wait and take your next dose at the regularly scheduled time.
  • Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light.

Drugs and Foods to Avoid:

Ask your doctor or pharmacist before using any other medicine, including over-the-counter medicines, vitamins, and herbal products.

  • Some medicines can affect how capivasertib works. Tell your doctor if you are using desipramine, efavirenz, erythromycin, itraconazole, midazolam, raltegravir, rifampicin, or verapamil.
  • Do not eat grapefruit or drink grapefruit juice while you are using this medicine.

Warnings While Using This Medicine:

  • This medicine may cause birth defects if either partner is using it during conception or pregnancy. Tell your doctor right away if you or your partner becomes pregnant. If you are a woman who can get pregnant, your doctor may give you a pregnancy test before you start using this medicine to make sure you are not pregnant. Use an effective form of birth control to keep from getting pregnant during treatment with this medicine and for at least 1 month after your last dose. Male patients who have female partners should use effective birth control during treatment with this medicine and for at least 4 months after the last dose.
  • Do not breastfeed during treatment with this medicine.
  • Tell your doctor if you have kidney disease, liver disease, diabetes, or a history of skin problems.
  • This medicine may cause the following problems:
    • High blood sugar level
    • Serious skin reactions, including erythema multiforme, palmar-plantar erythrodysesthesia, and drug reaction with eosinophilia and systemic symptoms (DRESS)
  • This medicine can cause diarrhea. Call your doctor if the diarrhea becomes severe, does not stop, or is bloody. Do not take any medicine to stop diarrhea until you have talked to your doctor. Diarrhea can occur 2 months or more after you stop taking this medicine.
  • Medicines used to treat cancer are very strong and can have many side effects. Before receiving this medicine, make sure you understand all the risks and benefits. It is important for you to work closely with your doctor during your treatment.
  • Your doctor will do lab tests at regular visits to check on the effects of this medicine. Keep all appointments.
  • Keep all medicine out of the reach of children. Never share your medicine with anyone.

Possible Side Effects While Using This Medicine:

Call your doctor right away if you notice any of these side effects:

  • Allergic reaction: Itching or hives, swelling in your face or hands, swelling or tingling in your mouth or throat, chest tightness, trouble breathing
  • Blistering, peeling, red skin rash
  • Fever, chills, cough, sore throat, body aches
  • Increased thirst, hunger, or urination, weight loss
  • Severe diarrhea, stomach pain, dry mouth, red or black stools
  • Unusual tiredness or weakness

If you notice these less serious side effects, talk with your doctor:

  • Nausea, vomiting
  • Sores or white patches on your lips, mouth, or throat
